I guess it depends on what you mean by "making it run", short answer I'm gonna say probably not.
The boot loader sets up UEFI GOP with a 640x480x32 linear video buffer. In TempleOS, I allocate a 32 bit frame buffer *gop_fb and patch the raw char routines to call UEFIGopRawPutChar which plots the character to the frame buffer instead of vga_alias.
To update the display, in TempleOS I call UEFIGopBlt which effectively does
Gop->Blt(Gop, *gop_fb, EfiBltBufferToVideo, 0, 0, 0, 0, 640, 480, 0); in the UEFI application.
So, under UEFI it is "running" with a 32 bit color display. Everything stays 640x480x4 (16 color) internally with TempleOS raw and Gr routines, and gets translated to 32 bit color value when writing to the display.
Could someone write programs to use different screen modes and 32 bit color depth? Of course. I won't be doing that, but there's nothing stopping someone from doing so.
Now, "making it run" - if I understand this correctly, "Is there any chance of someone coming along and rewriting all the code for WinMgr and every TempleOS app that Terry wrote to make them resolution independent and take advantage of enhanced color depth" it is possible but unlikely to find someone to fill those shoes.

Keep up the good work Alec. Have you ever considered making some sort of info-graphic or write up about the internals of TempleOS for people who really want to dig deep into the system? I know that the old website provides plenty of information, but a quick rundown could get people and developers interested into TOS who are just here for the Terry memes. I think too many people don't get the dream of having a x86 C64. Shit, just the idea that HolyC is able to execute code at compile time and then inject it back into the code stream makes my dick rock hard. We need to find a way to get people to appreciate TempleOS for what it is. I want it to be more than just a meme.

I dont know how the 16 color are used in TOS, but there is a way to emulate 24bits of colors
using only cyan magenta yellow and black with a pattern and dithering, it will reduce the resolution... see pic
The data, the dithering picture can be saved : 1pixel for 1bit in Black&White, processing it back with the pattern to restor cmy and restoring the colors.
