For most files virustotal.com is enough, if the file is big and I'm not sure for various reasons or too lazy to open a virtual machine I would just use windows defender or malware-bytes which is always installed.

Kaspersky Free on Windows without web protection, encrypted connection scanning, without using KSN (botnet), etc.
Glasswire FW Free version (free is not an ideal variant as a fw).
On Mac, common sense and Little Snitch FW.
I probably don't need an AV on Windows, but that's just in case I run untrusted or unsigned software or the trusted/signed software gets infected at the dev site.

Security Essentials on Server + All Client PCs. My desktop, where I do 95% of all internet browsing, I use just a standard user account for daily stuff. So no programs can be installed/changes made to the system without administrator creds.
