Spider-Man is a terrible match up for Deadpool.
When you make a comedy duo one of them needs to be the straight man. If it's kooky and quippy there's no comic-dynamic, there's no contrast.
That's why Cable works so well when paired up with Deadpool. He's a really ridiculous character but he's usually played very seriously. So he fits the tone of Deadpool in terms of being a big weird looking cartoon character but is a foil for Deadpools hijinks.
Rernolds and Miller get that which is why they had a very straight laced, boyscout Colossus in the first film.
