I'd say Lars and the Real Girl, even though the ending is happier. First it starts off with just an awkward shy guy who orders a sex doll and pretends it's real, but as the movie goes on you learn why and it's kind of sad.

The show M*A*S*H starts off pretty funny with lots if zingers and tomfoolery and eventually Hawkeye just sits around crying about a dead chicken.

Fun fact, the show lasted like 8x longer than the actual war. So I guess a turn to the dark was inevitable.

Doesn't really turn dark or sad as it goes along so much as there's a lot of dry, dark humour throughout and then suddenly the last 10 minutes of the series happen.

I remember Patch Adams hitting me in the gut when I saw that years ago though I don't remember why.

Devils at the doorstep

Random Chinese farmer is tasked by the communist to take care of two Japanese prisoners without the Japanese army (camped near his village) take notice of them. Hi-jinks ensue as the guy and his friends tries to find various ways to kills the prisoners without directly doing it.
It's genuinely funny in a fucked up way.

Then the movie takes a very dark spin near the end and you stop laughing.

Seems every year I forget it's about Thanksgiving and save it for a Christmas viewing.

The Invention Of Lying comes to mind as well until it turns into typical Gervais atheism.
