A quick search of the analogue shows that it reached stage 2 clinical trials, but no results were posted, which means it likely failed.

Drugs fail for a massive variety of reasons. Stage 2 trials generally look at the efficacy of the drug.
The patent for the drug was also probably filed in 1996, meaning that by 2009, they only had ~8 years left of patent protection, without even having concluded trials yet. Economically it was probably better to move to a newer drug.

You will have to recrystallize really well to make it stop being irritant to the eyes.

A tertiary amine NR1R2R3 can be further alkylated (R4-X) to give a quaternary amine salt R1R2R3R4N+ X-

You could create an amide when you use a carboxylic ester with an amine, but not in this case. That works only with ammonia, primary or secundary amines.
