You can but I don't see why you should. There are plenty of western comics that blend Japanese artstyles with western ones that look nice, why not try doing that yourself? Come up with your own blend and release something here that might get picked up in Japan as well.

Like any artistic endeavor, the problem is getting recognition in the field so that people want to publish your work.
You're competing with people who speak the language and are native to the culture, so unless you have amazing skills there's no reason why your work would get singled out before any of the thousands of native Japanese would-be mangaka.

You don't have to be Japanese but you need to be pretty fluent in the language. There are even college exchange programs to get into drawing manga if you know enough Japanese to live there.

If you make a work, it will sell at least some.
Practice every waking moment and you will publish; literal children publish manga and LN's in Japan.
If your manga has a distinct 'japanese' aesthetic, story, and characterization the Japanese publishers will publish the work.
Publishers only care about one thing "will it sell", it does not matter if your art is bad and your stories are swiss cheese...all that matters is making them believe that it can sell.

I'm struggling to fill just one book. It's my first piece of creative writing I've done outside of school and my pacing is full retard. MC already wins over her love interest and achieves the goal she and her best friend set for themselves setting up a good stopping point for the story. I feel like the second half can only be finished by going needlessly dark with MC and her love interest breaking up and MC sleeping around and developing an alcoholism problem and her love interest attempting suicide at some point. But I feel really bad because I'm attached to the characters and I can't bare to make them suffer.

Mate, that sounds awful. While it would be a change of pace from the usual dragging out, it's just too fucking quick to let people get to know your characters. It would feel like two random people get in love, told by a third party. No details, or if there are, it will be info-dump storytelling, where people say everything about them to eachother outloud, without leaving any mystery.
I also really doubt that the average person on here would be able to write believable dialogue and characters.
That needless darkness you want to do, is really fucking dumb, as well. I suppose that I won't be able to grow as attached to them as you, and would not really feel any sympathy for them. Suicide? Alcoholism? You're making them sound rather gratuitous, without any real substance behind them. With possible flash-forwards? If you seem intent on doing it, then I see multiple way you could do it:
>sudden, abrupt flash-forward, with flashbacks (that would be touching in animated form - with bright colours, bloom, chromatic aberration and the like)
>multiple, short flash-forwards - really leaving no room for sympathy here, unless you can do it well, and give each part at least 30-40 pages
>darkness all the way, with one 10-page flashback; with a sequel novel exploring everything that went wrong in the past in detail, half from the MC's perspective, and half from her love interest's (interwoven parts, not half-half). The third volume would also explore both, but on their way to a new life - not necessarily with eachother.

But really, I feel like your thing would work best as a WN. If you do manage to find readers, you'll be able to pander to them to a certain degree, or do the exact opposite. Don't do that constantly, though. For example, if most readers want to see two background characters (you did mention some of them now, right?) fall in love, by all means, do mention them occasionally - so you can bait them have them at your mercy. If you want, make them angry - that works really fucking well, because only things like that cause shitstorms, and thus you get more readers because of the increased popularity; or pander to them. It's completely up to you. Don't even go that route if you don't want to.
