Nui and Ragyo didn't see themselves as human so they looked down on Satsuki and Ryuko as a lesser species. Only after Ryuko was revealed to be the same type of "creature" as Nui and Ragyo did Nui start treating her the same.
Also Ragyo considered Nui a failure since she couldn't wear a kamui, so she probably raised her to believe she was just a spoiled murder tool.
Nui was definitely at least part human, as life fibers by themselves could not take on a human form. Kamui was the most advanced purely life fiber form, but even then it required a human host. Life fibers by their very nature were symbiotic, so they had to have a human host. I don't see any reason to believe Raygo just grabbed some random sperm and egg from two other people before sticking it in the oven, so I'm sure she'd continue to use her own body to experiment on as she had previously.

Note that when infant Ryuko "died" she complained that her genetic material wasn't good enough. Combine that with the fact she had REVOCS science colonies in at least Africa (Rei backstory) looking for suitable subjects for Life Fibers it's entirely possible she did just that after combing through millions if not billions of possible test subjects to pick out the potential best candidates.
