I'm sorry but no, criticize the heroes all you want (and not all heroes are like what you described), but they still make the world a better place and help people. All the villains do is complain about society is unfair because they don't accept their criminal ways and then proceed to make society worse by destroying, killing and stealing.
That's like saying that because some police are sometimes too brutal or act like jerks, that means that gangsters, murderers and rapists are better.

Never ever. They're only 30% of the population, most of them are probably old, and they have an inherent disadvantage in an actual combat situation.
But really though, why would there be an uprising? In Japan at least, Quirk use is heavily restricted, and for most jobs having a Quirk doesn't seem to matter. Other than getting bullied at school and the inability to become a hero, it doesn't seem like non-Quirk users are getting the raw deal, life continues as normal.

What about someone like Knuckleduster, who could clearly pass any physical qualifications and is a talented enough combatant to dispatch villains in unarmed combat? There's plenty of examples of people at UA whose quirks take a back seat to their other qualifications. Eraserhead is as good a hero as he is not because of his quirk, but because he's a damn good fighter. Mei's quirk basically does nothing for her whatsoever, it's the fact that she's got a genius IQ that's gotten her this far.

Motives that actually make sense, a real feeling like their actions are not justified but are accord to the character, an interesting and well done backstory and being the least possible of a cokie cutter character.
Something that doesn't feel like fan service or hamfisted shit.

Or Nighteye, his Quirk is actually not very useful in most combat situations, especially since he can only use it on one person (that he has to touch) once a day. And he doesn't even use any weapons or a special suit.

Well, it depends if you prefer to watch the anime than read the manga, but the anime has actually been very faithful to the manga, only expanding some things, like the fights. I personally like the manga better, so I recommend it

In Eraserhead's case it's less a matter of his own skill and more of a matter of how dependent others are on their Quirks.

Knuckleduster, on the other hand, has never faced off a top or even mid tier villain before so it's probable that his capabilities are ultimately limited as much as he trains himself.

More than a Quirkless uprising, what I could see happening is a non-hero rising. All the civilians who can't freely use their quirks, unlike the heroes, and all the people with quirks that are pretty lame that they might be quirkless, or with dangerous quirks that are automatically feared. Quirkless people is on the way to extinction, though, most of the quirkless are old people and a kid being born without a quirk is extremely rare. BnHA is Magneto's wet dream come true.
