It's a question of whether that's enough to wipe out all of Herakles' lives with one hit. Using Knight of Owner on something that isn't a Noble Phantasm will only result in a D-rank weapon at best. Herakles wouldn't even feel that. His A-rank strength would be able to pierce Herakles' skin, but if he fails to take a life, God Hand will gain a resistance to it and Lancelot would have achieved nothing.

It all boils down to whether Lancelot can wipe out all of Herakles' lives in one shot or not.

Knight of Owner only affects that which is conceptually a weapon. God Hand is a defensive Noble Phantasm, so that concept overrides any remote possibility that it could be considered a weapon.

Arondight can't shoot beams. Overload isn't a beam, but a charged slash. Also, even if they have the same rank, do recall that Excalibur is explicitly the most powerful of all Holy Swords. Don't assume Arondight is its equal just because they're sister blades. Caliburn is selectively A-rank under the right conditions. It was able to wipe out seven of Herakles' lives in one shot. Plus, there's no guarantee that Herakles won't pull his "surpassing his legend" asspull and throw out one more strike even after all his lives are exhausted.

Frankly, the match is still in Herakles' favor. Herakles has twelve lives. Lancelot only has one. The battle will only become more and more disadvantageous for Lancelot the longer it drags on. In order to have any real shot at victory, he needs to end it as soon as possible.
