I think Daisy should have her own slot because I feel that Peach's attacks don't suit her personality bery well, plus I want a Smash series Daisy amiibo and for her not to be overshadowed again. Daisy won't ever be a model swap for Peach for the same reason Lucina wasn't one for Marth: she's noticeably shorter, and it's not something Sakurai is likely to ignore. It's also important to note that Smash Peach is proportioned differently to Mario Peach: the latter is more like an 8-year old, while the former is closer to a 12-year old. Give Daisy the proportions of Smash Peach, and it just doesn't look right. Pic related comes from a Daisy costume mod for Peach in Brawl: there's nothing wrong with it, per se, but she looks huge compared to her Super Mario look. It's just too tall and doesn't look right.
As for making her a unique playable character, I don't think so. She doesn't really have any specific abilities unique to her that could make for an interesting fighter. People have talked about adding her as the "Spin-off" character, her move set incorporating various references to the different Mario spinoff titles. If they ever decided to make a character like that, then it would have to be Waluigi. Unlike Daisy, he actually debuted in one of these games, so his identity much more closely connected to them. He's also quite visually distinctive, his comical personality and mannerisms lend themselves better to a crazy move set like that. You can easily imagine him running down the stage in this hilariously over-exaggerated manner, throwing Dice Blocks or serving tennis balls at people as he whines and "WAH's": he lends himself better to it than Daisy, and it would probably be more fun to watch and feel more fun to play. He's also more popular.

Waluigi and Daisy are without a doubt the next most prominent characters before you dip into generic enemies, except maybe Baby Mario & Luigi or Paper Mario.

I could totally see them adding in a "retro" character that specifically represents the original arcade Donkey Kong games, fighting similarly to how Pac-Man and Mr. Game and Watch do. But that character would have to be Donkey Kong Junior.
