You can either have this decent joke or have them talk at each other in awkward stiff language with a designated straight man for every single joke ever.
Apparently japanese people are physically incapable of identifying a joke without a character that does nothing but yell at the audience that a joke has just occured.

Seriously, all Japanese comedy is either that straight man and idiot routine or stupid puns involving kanji looking like other kanji.
Well, then there's the "comedy" of an idiot main character accidentally tripping into a girls tits and getting punched, but I don't think even Japs really think that's funny.
They can make some really funny slapstick comedy, but even that tends to follow a very rigid framework.
I do like me some rakugo, though, even though it is the very epitome of structured comedy.
It depends on the media. I find Steins;Gate an interesting case study here. The official translation of the anime is hot garbage because they went down the path of pandering to reddit, replacing the original Japanese imageboard memes in the process. The VN on the other hand had a pretty much perfectly fateful translation which is brilliant, but the memes and other cultural jokes are only understandable because of an in-game glossary that was also present in the Japanese version.
My point is that there's no right answer and the solution is a competent translator that knows when to play it straight and when to replace things.
I can think of a few cases where their humor consistently translates well beyond slapstick, but they're rare.
