To elaborate, humanity is linked with physicality, and souls with the ethereal. By extension, humanity is linked with matter, (hence dark magic being described as "weighty") and strength, whereas souls are linked with intellect and magic (hence the fact that most sorcery is apparently derived from the soul).

As for why one goes hollow when they lose their humanity, it's simply because humans are a kind of pygmy, and being hollow is their natural state. It's only the presence of humanity that blesses pygmies with the reason a soul can grant.

A GAME THEORY

Well it's safe to assume most skeletons were human at one point, and the rats in the depths eat a lot of corpses. Bosses I have no idea why.

In the beginning, born out of the Flame, there were four Lord Souls: Life, Death, Light, and Dark. Humanity is linked to the Dark Soul, which was found by the Furtive Pygmy. The Pygmy hid his Lord Soul from Gwyn, Izalith, and Nito by dividing it up and giving it to his offspring, humans. Hence the name "humanity."

Souls and Humanity are linked to the Fire, which represents the power to change reality, and also entropy, which creates the arrow of time, so that things don't remain static. Thus, the more souls you possess, the greater your power, because power would have no meaning in an eternally static, unchanging world, you see.

TL;DR, the only real difference is aesthetic, since each piece of humanity represents a shard of the original Dark Soul, they look dark. Metaphysically, both represent power, however, humanity is tied to darkness, so it is symbolic of the shadow cast by the Flame.

Addendum: you might also speculate that the Dark Soul was different from the Light and Life Lord Souls, in that - together with Death - it represented the inherent entropy of the Flame, since Fire requires fuel to burn, and therefore implies that the world ages and will eventually fade back to the static unchanging reality it was before the beginning of time.

I heard somewhere that that dark soul was different in that it continuously multiplied, which is why there is still so much humanity lying around in the first game.
