You'll miss some returning characters and plotlines but the story should do it's own thing, the main three are all new characters.

I'd say you should at least grab the first game's OST though, not only is it simply beautiful and the best part of Nier 1 but the composer is returning and remixing some of the first game's tracks in Automata.
