There are anons with theories about that Clancy was going to be the protagonist of the main game but because the voice actor passed away, and that Japanese people respect v.a. too much and they don't like to recast roles, the main game's plot was re-written with Ethan as the protagonist and Clancy dying.

Not necessarily, since the DLC could quite easily be set during the later events of the main game. Event flags in the latest patch suggest that the premise of Not a Hero is Redfield tracking down Lucas to either take him into custody or kill him, and we know from the radio transmission in the salt mines that they're still keeping an eye on him. So there's a window you could easily place something into.

In the Daughters DLC she seems to have the bug ability instantly. This is a little far fetched and completely unfounded, but my personal theory is maybe she already had a parasite of some kind, I mean they live right on top of a shitty bayou.

Serum created with D-series material is only somewhat effective on an E-series infection. Mia wasn't completely cured of her Mold by the serum, only enough to resist Eveline's control and allow Ethan to escape the tanker.
