Only if you use lossy compression. For example, PNG is a non-lossy compression method (literally a zipped BMP that's unzipped on the fly) and JPG is a lossy one.

Uncompressed anything is a meme. Only thing you lose by compressing image data is a miniscule amount of processing power (irrelevant for a 2D platformer that has barely any demands on the processor anyways) and only thing you lose by using variable bitrate MP3 instead of FLAC for audio is nothing, since human ears can't hear the difference anyway.
