Are there any adventure/single-player RPGs where being a bare-handed martial artist is a viable option? (At best equipping gloves, special gauntlets, etc.)
God Hand's combat system is well-vaunted to this day, and there was Jade Empire way back when, but nowadays bare-handed combat might as well be "tutorial" animations until you get the sword or whatever that you're actually expected to deal damage with.
Best you can get is modding something like New Vegas or Dead Island for barehanded melee to be less pathetic compared to guns and things.

In Vampire The Masquerade: Bloodlines you can do a pure fist build with most races, Brujah probably being the best, or a more "transform into a beast with claws" build with Gangrel.

Usually warrior games have at least 1 character or weapon which represents punching. At the very least the dynasty and samurai ones have since they cover a lot of weapons (gauntlets), but some spin off games should too.

Divine Divinity 2 (older third person RPG, not the relatively new isometric RPG) has very viable monks/martial artists. Oblivion is so-so, your max damage is pretty low (11-12 IIRC) but you attack pretty fast and do fatigue damage, plus it changes the whole game into more rhythm-based combat (dart in, punch them 2-3 times and backstep before they can retaliate) unless you want to abuse paralyze and magic.

If you want to mess with first person brawlers there's Zeno Clash, but I couldn't get into it all that much. Absolver also came out recently, dropped that as well for some reason. Borderlands has Brick I think who's active skill was a berserk fist button. Chivalry had unarmed combat which was kind of funny, but I haven't played that game in years. From what I've seen the PvP looks ridiculous now. DMC games have Ifrit/Beowulf/Gilgamesh. And finally I guess Sleeping Dogs also has some BamHam combat.

How could you do a kind of a pro fighter/street fighter/brawler kind of class?
Characters derived from the character archetype of the Shaolin monk often use ki attacks and other mystical stuff.
In Pokemon, there's the Dark type, where everything is kind of themed around desperate and dirty fighting. e.g. compared to the Fire/Fighting types, Incineroar is designed as a heel wrestler, and has signature moves involving attacking the throat and lariats.
In Etrian Odyssey 5, the pugilist has abilities around striking enemy's limbs to prevent abilities being used.
I mean, it can be as simple as most other damage-oriented classes(e.g. element punches, self-buffs, things) but how would you theme it?
Self-anesthesizing, like focusing around taking painkillers or drinking booze during a fight for self-buffs? Or self-heals that look like you're acting as your own cutman?

Thing is, a lot of games have the brawler tied in with berserker, which I never really liked. Meaning damage increases as health decreases.
But it's either than or make a dodge monk, which is cool too, but never satisfying in turn based RPGs. Beat em-ups and brawlers always did it best, where you could actually have an active fight and dodge/block/parry moves.

It's literally just a question of animation. Drop the finesse and add in heaps of brutality.
As for class features, I think some sort of damage soaking (ignore pain type of deal, maybe?) would fit the best.
