You starting on a planet with few resources and a "you WILL make this inhabited planet very profitable for the emperium". Fuck ALL it would need would be a good terrain generator, unit implementation, random event generator and so on. A lot cheaper than the dogshit garbage that was DoW3.
You'd start small, have to get resources locally and slowly make the entire world important enough to even requisition a Space Marine chapter's help as a sort of "hero unit".
While rimworld has big problems with the random events, I think with a basic RTS unit control like the one on DoW you could balance it better, depending on the startup settings you could select which types of enemies you'd find, the types of events, which resources would or not be available on the planet, the availability of trading with other planets for resources you didn't have and maybe even some focused objectives for the game, like turning the world into a forge world or a farming world or a training world. Having to balance things like city morale with the proper buildings, having to manage the likelihood of chaos cultists appearing and being successful with investigating units, it's full of possibilities.
All those narrations of walls of lasgun shots being a work of art and you'd be able to create that yourself. Imagine a random event of a warp storm where you have to defend the planet/city against a chaos invasion while being cut off from Imperium's help.
Hell maybe it would even be possible to pick a planet in a specific area of a galaxy map to influence your chances if you don't feel like controlling every aspect of the settings, maybe even pick famous worlds such as Cadia to play through a very specific scenario.

The nids aren't of anything, except for something.
It'd be less about tower defense and more about long term RTS/city building. See Eldar would be able to pass right through your outer walls if you didn't had the proper defenses, and chaos cultists appearing from within your ranks would be able to warp Chaos right into your big city if you didn't stop them before that.
Also Ork ships and nid spores. You'd be forced to manage some actual city defense with your troops.

The Imperial Guard generally doesn't give a fuck about all that when the higher ups aren't looking. They fix their own gear, use the uplifting primer as Toilet Paper, and in Catachan's case they frag Commissars.
