It's obviously not practical by any means, to do it by hand.

Can you? Yes. Assembly is just a set of mnemonics for machine code, so any application which can be expressed as machine code can also be written in assembly.

Should you? Hell no. You won't gain a significant performance boost, but you will spend more time programming, and you'll lose portability.
