1. The Jedi are going to test every single Senator when it could be none of them, it could be a member of a senator's staff, it could be an infiltrator, it could be someone who was not a senator.
2. How do we even know the Jedi have the time, people and resources to take a blood test for tens, if not hundreds of thousands of people? Even if they had all of those, it would still be a time consuming process, and would also alert the Sith and give them enough time to entrap the Jedi.
3. What if a Senator has an especially large amount of Midichlorians? It is highly possible a senator's race may be particularly prone to having a high Midichlorian count, Yoda's race, for example?
4. Having a high Midichlorian count does not prove that you are not a sith, it just proves you have a high affinity with the force. This would also potentially put the Jedi in hot water if they wrongly convict someone, and makes their position weaker against the Sith conspirator who seeks to undermine the Jedi.
5.What happens if enough Senators don't take the Jedi seriously? The Jedi may be guardians of the Republic but it's possible quite a few systems do not have much respect for the Jedi and their Senators would not care to comply with the Jedi's request, just refusing to have a Midichlorian test is not enough basis of evidence for the Jedi to suspect someone of being a sith when there could be many other reasons.
