That's actually how I played the first time, because I'm a huge autismo.
My favorite "challenge" is to RP. In every Divine Beast region, you must explore/complete the dungeon with the armor and weapons of that region.
Other RP locations include the Sheikah set/weapons for sneaking into the Yiga base and the Knight set for storming Hyrule Castle.

no using the map ever, no fast travel and you must guide yourself with landmarks only
The "no teleports" seems kinda arbitrary, especially for quests that require you to travel across the map. For the Zora wife quest, are you gonna run all the way to Lake Hylia and all the way back?
I think Link's initial inventory size is properly balanced. Increasing it kinda breaks the game.

It's a bit of a bitch, but it's how I always play my open world games, dating back to Oblivion. But then again, I just like walking around and finding stuff.
I think limiting teleports is better than ruling them out
Like only teleporting to towers, not shrines, seems like a better approach
In that case, paragliding should be discouraged
You're not really finding much on repeat play throughs, especially if you've already 100%. It's a nice rule for an initial run.
What if you made a no stamina upgrades rule
Well, that's how I played my initial run. I haven't played since, since I'm waiting on DLC for a Hard Mode Run.
