Sorry, nu-Sup Forums believes that self-imposed challenges are bad game design. If there is something optional in a game that you dislike using, the game is shit.
I think the only times I've really done something similar are the Mega Man X games on the snes. Buster only/no upgrade runs are fun and really change how you play the game.

sl1 dark souls is the only challenge run i ever did. it was fun.
Beat Fallout New Vegas without leveling Survival or Medical.

Last one I've done was a Mass Effect 2 play through where I don't invest talent points and use no weapons or armor besides the base ones.
My favorite playthroughs are going through both Bioshocks without spending ADAM or PttP machines, using the Research Camera, and I have to provoke every Big Daddy I encounter.
Dark Souls, at soul level 1, no shields, no summoning, no pyromancy, caestuses only.
Morrowind:
No alchemy abuse, no stealing (unless needed for a quest), no training, no fast travel until discovering the place, no fun
Been wanting to do a run of final fantasy 5, rngesus run. Literally the only things I'm allowed to use are actions that have a random/ever-changing. Effects of outcomes, like the deer class or dancer, beserker is also allowed, along with use of items like the wonder rod. I can grind up and use other non random stuff like dual wield and the rapid strike, (i wanna dw wonder rods with rapid strike just for shots and giggles). Passive stuff that heals is allowed, only because rng self healing is likely not going to work out well, but I can't use items unless they have a variable chance affect, nor can I apply buffs unless the source of said buff is rng based. Armor/equipment is the only reliable buff scources I'm allowed

I never made it past there. It was just too much bullshit for me to handle.
I never up survival, but medicine I usually put around 30-50, for certain perks
Hell why even stop there?

Buster Dueling the first 8 bosses in any Mega Man is the true way to play.
Its not too hard either outside a few fights like Shadowman.
It's unbeatable then, you need the baby yoshis eat move to be able to beat one of the arena fights.

This was massively fun in MGS4. My Snake became a force of nature. MGS1/2/3, not so much. Especially 3. Haven't tried it in Ghost Babel or the PSP games yet, and don't have V.
Fucking masochist. I'm only on my second playthrough, doing a no physical attack run, and good LORD is it brutal. Thank God for Heavy Soul Arrow, Lightning Spear, and Fireball.
The knife didn't get seriously nerfed until after Code Veronica, so that's a pretty good challenge. I did it in DS, tried and failed in 2. It's particularly easy in 4, though, because you can just knife people in the face then kick them, except for bosses.
I'm on my first playthrough and doing that just because ammo is rare and guns tend to suck anyway. Did I do something wrong with my SPECIAL points, or is early game just like that?

I feel like nuzlocke is a bit boring but I often play Pokemon only catching and raising dudes that are below a certain stat total or make 'wrong' builds for the same reason such as special moves only on physical dudes or monotypes
the truck team at the wal fart I work at is doing a nuzlocke run where they pass the game boy to a different person every day, even the supervisor is in on it

A few years ago I beat Super Mario 64 100% with only my feet.
There's still plenty of bullshit after that. After that you have to go through Silent Prison, and some retard at the controls sped up the guillotines to absurd amounts. It's at the 5:45 mark.

Nuzlocke in one of the Pokémon games or hacks. Probably FireRed, HeartGold, or a FireRed hack.
It was interesting to see how these otherwise fairly easy games became much more challenging. Standard rules of "one per area" and "if it faints, it's dead".
