When I was a kid I thought the episode of Ed, Edd, and Eddy where they turn Johnny into the most annoying creature on earth was the most hilarious thing ever.
I was rolling on the floor for the entire episode.
I remember a good portion of EEE spilling my guts with laughter.

I never laughed at this though, I only ever masturbated to it.
Not sure if hardest, but the earliest I can remember a toon making me ROTFLMAO was the "What a Cartoon!" short "Strange Things".
Something about the rapid cuts and zany sound effects, which I later also found amusing in YTPs, coupled with a dissonance between the silly action and somewhat gloomy early 3D lighting.

The first time I watched this I was 14 and I was laughing so hard I started gasping tryingto catch my breath. By the end I was holding my sides in agony they were so worn out from the laughter.

I must've watched the first 2 seasons of american dad like 30 times, i've never bothered with the other ones, dey good?
It is hard to say because regulary I have good laugh because of subtle or even pedestrain jokes. For example I laughted really hard when Linda from Phineas & Ferb waiting on the phone and listening to chip tune version of her own song. Or when in Clone High after Joan of Arc demonstrated her movie Freud said something like "oh, somebody in love!".
But yeah there are also stuff like pic related and it got me from time ot time.
