What if the reason you think that those with great power misuse it is because most people with great power ARE actually responsible and you only hear about the irresponsible use of it? Also Spiderman's response can be taken as sarcastic to such an inane statement.

I can understand people having issues with batman's no kill rule or something like that, but spidey's motto is a good thing to live by, no matter what level of power you have. I think the point she's making is that being powerless doesn't excuse you from responsibility. You don't need powers to be Superman, so just because you don't have them doesn't mean it's okay to not do good.

It's worded in the dumbest possible way though, and the whole "keeping them in check" part is totally worthless. If you have 'powers' then yes, more responsibility applies than the average person would have because you possess a greater capability to help the world.
